Pyrroloquinoline Quinone as Cofactor in Galactose Oxidase
Galactose oxidase from Dactylium dendroides was shown to contain one molecule of covalently bound pyrroloquinoline quinone (PQQ)/enzyme molecule. From the spectroscopic characteristics reported for the enzyme forms, a mechanistic role for PQQ could be deduced. In analogy with other quinoproteins, the initial formation of a PQQ-substrate adduct is proposed. Following internal hydrogen transfer, ...
متن کاملPea (Pisum sativum) diamine oxidase contains pyrroloquinoline quinone as a cofactor.
Diamine oxidase was prepared from pea (Pisum sativum) seedlings by a new purification procedure involving two h.p.l.c. steps. We studied the optical and electrochemical properties of the homogeneous enzyme and also analysed the hydrolysed protein by several methods. The data presented here suggest that the carbonyl cofactor of diamine oxidase is firmly bound pyrroloquinoline quinone.
متن کاملReactivity of a functional carbonyl moiety in bovine aortic lysyl oxidase. Evidence against pyridoxal 5'-phosphate.
Previous studies have pointed towards a cofactor role for pyridoxal 5'-phosphate (PLP) in lysyl oxidase, the enzyme that generates the peptidyl aldehyde precursor to the lysine-derived cross-linkages in elastin and collagen. The nature of a carbonyl moiety in purified bovine aortic lysyl oxidase was explored in the present study. A PLP dinitrophenylhydrazone could not be isolated from lysyl oxi...
متن کاملReactions of the oxidized organic cofactor in copper-depleted bovine serum amine oxidase.
A novel copper-depleted bovine serum amine oxidase (BSAO), in which about half the molecules contained the organic cofactor in the oxidized form, was prepared by adding a reductant in anaerobic conditions to the cyanide-reacted protein. The CuI-semiquinone formed in these conditions reoxidizes after the removal of copper. The inactive derivative was reduced by benzylamine at approx. 1/1000 the ...
متن کاملPyrroloquinoline Quinone a Redox Cofactor and Its Involvement in Biological System
This paper highlights some features, which describe the characterization and physiological importance of Pyrroloquinoline quinone (PQQ), discovered more than forty years ago. Mechanism of biosynthetic pathway of PQQ is described based on sequence analysis and homology models. But still now all activities of PQQ are not discovered. PQQ is not only a bacterial redox cofactor in alcohol oxidation ...
